Lozam 2 Mg 40 Tablets (lorazepam)

Studies in healthy volunteers showed that a single high dose of LOZAM® had a tranquilizing action on the central nervous system (CNS) with no appreciable effects on the respiratory and cardiovascular systems. LOZAM® is readily absorbed with an absolute bioavailability of 90%. Peak plasma concentrations occur approximately 2 hours after administration. The maximum plasma level for 2 mg Lorazepam is approximately 20 ng/ml. The elimination half-life of unconjugated Lorazepam in human plasma is approximately 12 hours and that of its major metabolite, Lorazepam glucuronide, approximately 18 hours. At clinically relevant concentrations, Lorazepam is approximately 85% bound to plasma proteins. LOZAM® is rapidly conjugated at its 3-hydroxy group to Lorazepam glucuronide which is then excreted in urine. Lorazepam glucuronide has no demonstrable activity on the CNS of animals. Lorazepam plasma levels are proportional to the administered dose. There is no evidence of accumulation of Lorazepam in administrations of up to 6 months. Studies comparing young and elderly subjects showed that the pharmacokinetics of Lorazepam remain unchanged in elderly subjects.
