The active substance is manifested by a reduction in food intake and only to a small extent by an increase in metabolism. The site of action is probably the lateral hypothalamic feeding centre. In humans, tolerance to appetite suppression develops, so that sustained weight loss is not observed in obese individuals without dietary restrictions. Clobenzorex is absorbed from the gastrointestinal tract after oral administration of the product and is excreted in urine; one of the main metabolites is amphetamine (15% of the administered dose) and p-hydroxy-clobenzorex (1.5% to 6% of the administered dose). Clobenzorex is a sympathomimetic amine with a pharmacological activity similar to other drugs of this class used for the treatment of obesity. Its effects include some stimulation of the central nervous system and a transient increase in blood pressure. Weight loss in obese individuals treated with sympathomimetic amines is almost entirely due to lack of control of obesity.
